Dose response curves Flashcards
what is receptor based model
binding of drug to targets can result in response that can be modelled graphically
what is law of mass action
rate of reaction is proportional to [c] of reactants
what assumption of law of mass action
ligand binds in a reversible manner
what is fractional occupancy of receptor formula
[bound receptors] / [total receptors]
** key response formula
response = [drug]*[max response] / dissoctiaion constant + [drug]
what is ED50 on dose response curve
dose that give 50% of max response
what is efficacy
max effect a drug can have - ideal world
what is potency
reflects the dose/conc. required for specific effect
what values can be used to compare potency
ED50
what does higher potency mean
more effect with less dose
rule of thumb regarding doses
generally want to give the smallest dose possible
3 types of agonists
- full - prefers active state and gives full effect
- partial - has less efficacy when bound - never full effect
- inverse - cuases action, but reverses state
what is competitive antagonist
competes for binding sites and shuts down agonist action
how to overcome comp. antag
give more
2 types of non-competitive antagonists
- irreversible - binds and never lets go - can’t overcome
2. allosteric - binds to different site, but changes the affinity of binding site to agonist
