CC LAB - TPAG Flashcards
Based on measurement of nitrogen content
Kjeldahl Method
Reference method
Kjeldahl Method
Digesting agent of Kjeldahl Method
H2SO4
Most commonly used
Biuret Method
Need two peptide bonds and an alkaline medium
Biuret Method
Composition of Biuret Reagent:
a) Copper sulfate- provides the cupric ion that complexes with the peptide bonds
b) Potassium sodium tartrate (Rochelle’s salt)- prevents precipitation in alk solution
c) Potassium iodide- prevents autoreduction of copper
- important in maintaining plasma oncotic pressure
Albumin
family of globular proteins that have higher molecular weights than albumin and are insoluble in pure water
Globulin-
Inverted A:G ratio means
presence of myeloma
Immediate response occurs with stress or inflammation caused by infection, injury or surgical trauma
Acute inflammatory response
- normal or ↓ albumin
* ↑ α1 and α2 globulins
Acute inflammatory response
Late response is correlated with chronic infection
autoimmune diseases, chronic liver disease, chronic infection, cancer
Chronic inflammatory response
- normal or ↓ albumin
- ↑α1 or α2 globulins
- ↑↑ γ globulins
Chronic inflammatory response
Cirrhosis can be caused by [2]
chronic alcohol abuse or viral hepatitis
↓ albumin
• ↓ α1, α2 and β globulins
• ↑ Ig A in γ-fraction
Liver damage - Cirrhosis
