acid-fast. due to waxy mycolic acid in their cell wall. gram’s stain indeterminate when we do grams stain despite having gram positive cell wall structure.

what is the primary acid fast stain?
carbolfuschin - basic dye, cationic
fix cell..
steam for 3-5 min. dye already has phenol.
mordant melts
mycolic acide cell wall (acid fast)
after fix..
cool slide for a few minutes carbon dye trapped inside cells. add decolorizing, no time limit. add counterstain, methalyin blue. wash off, dry, observe.
