PHENYTOIN Flashcards
1
Q
Drug class
A
anti-convulsant
2
Q
How does it work? (4)
A
- reduces neuronal excitability and electrical conduction with in the brain
- it inhibits the spread of seizure activity
- it does this by binding to Na+ channels and preventing Na+ influx into the neuron
- prevents the trigger of an action potential
3
Q
Indications
A
- seizures in status epilepticus
- one fit after another without regained consciousness
4
Q
Contra-indications
A
- hepatic imapairment
- dose reduced - pregnancy
5
Q
Side effects (7)
A
- changes in appearance
- skin coarsening
- acne
- hirsutism
- gum hypertrophy
- cerebellar toxicity
- impaired cognition or consciousness
- haematological disorders
- osteomalacia
- hypersensitivity reactions
- death
6
Q
Interactions (3)
A
- drugs metabolised by cytochrome P450 enzymes
- reduces plasma concentrations
e. g. warfarin - cytochrome P450 inhibitors
- plasma conc increases of phenytoin - drugs that lower seizure threshold
- efficacy reduced
e. g. SSRIs and tricyclic antidepressants
7
Q
Elimination
A
urine
