Propofol Classification

Sedative-Hypnotic
Propofol Mechanism
Medscape:
Short-acting, lipophilic sedative/hypnotic; causes global CNS depression, presumably through agonist actions on GABAa receptors
Capital:
Works on multiple CNS receptor sites including GABA to cause rapid anesthesia
Propofol Onset of Action
Capital: Immediate
Propofol Duration of Action
Capital: Effects stop nearly immediately after infusion is discontinued
Propofol Indications
Capital:
Anesthesia induction, Anesthesia maintenance, Sedation for mechanically ventilated patients
Propofol Contraindications
Capital:
Hypovolemia, Hypersensitivity to drug, soybean oil, peanuts, or eggs.
Propofol Side Effects/ Precautions/ Comments
Capital:
Hypotension, Seizure, Involuntary muscle movement, Nausea/vomiting, Dysrhythmia
Acute renal failure, Apnea
In pediatrics, Propofol can cause extreme bradycardia when administered with fentanyl
Avoid rapid administration in elderly due to high risk of hypotension
Propofol Adult Dose
Capital:
Induction: 1.5-3.0 mg/kg IV/IO
Maintenance: 25-100 mcg/kg/min IV/IO infusion
Medscape: 2-2.5 mg/kg
0.1-0.2 mg/kg/min
Propofol Pediatric Dose
Capital:
Induction: 2.5-3.5 mg/kg IV/IO
Maintenance: 125-300 mcg/kg/min IV/IO infusion
