What are the steps in a Gram stain preparation?
- Mount the specimen on the slide
- Stain with crystal violet
- Use iodine as a mordant so the color isn’t washed out
- EtOH/Acetone wash (Gram + will stay violet)
- Counterstain with safari (Gram - will stain pink)
What are the best specimens for a direct exam?
- Sputum
- Urine
- CSF
- Vaginal Smear
What are Acid Fast stains used for?
They are used for bacteria with cell walls of long chains of fatty acids
What are examples of bacteria that Acid Fast stains?
- Mycobacteria
- Nocardia
