  1. Progesterone receptor antagonist- binds to the progesterone receptor with a higher affinity and there is not down stream signalling
  2. Only licenced drug for termination of pregnancy in the bitch. Termination of pregnancy at any stage

Dose rate: 10mg/kg = 0.33ml per kg twice 24hrs apart.
3. Injection site reaction

Anorexia, excitation, depression, vomiting, Diarrhoea.

If on glucocorticoids it can reduce their effects (acting as an anticorticoid) but she will only be administered the drug twice

Oestradiol-3-benzoate/ mezalin

  1. Action?

A
  1. Prevents implantation and/or interferes with transport of zygote.may continue to show signs of oestrus- can be mated again although they won’t become pregnant.
  2. Mezalin is unlicenced and off market. Use on day three, five +/- day seven post mating.

Dose= 0.01mg/kg given on day 3 and 5 after mating (day 0). A third dose on day 7 if the bitch has mated several times or if we don’t know when the mating was. Sub, IM
3. Overdose possible.

Narrow therapeutic index.

Fail rate of 5% i.e. 5% of bitches remain pregnant.

Can lead to pyometra and can lead to bone marrow suppression.

Can result in alopecia and hyperpigmentation

Prolactin inhibitors

Galastop

  1. Action?

  1. Selective and long lasting inhibition on prolactin secretion. Treatment for false pregnancy in bitches and suppress lactation
  2. Dose rate: 0.1ml/kgbw once daily for 4-6 consecutive days depending on severity of clinical signs.

Offlicence to use for pregnancy termination.

Efficacy of 80-100% for treatment of false pregnancy
3. Contraindicated in pregnancy because it causes abortion in bitches in the later stages of pregnancy but this may be desired

Prostaglandin and prolactin inhibitors together

  1. Cabergoline (galastop) at 5mg/kg per day for 10 consectutive days combined with

Coprostenol (estrumate) at 2.5mg/kg every second day on five occasions (every other day)

Off licence use of this formulation to terminate pregnancy

Aside from making the assumption that Cleopatra (unwanted mating brought in by the owner) has been mated and has been mated within the fertile period: are there any methods that you might use to propose mating has occurred and whether risk of pregnancy was low or high?

  • Saline moistened swab placed in vagina for one minute
  • Swab tip cut off and placed into 0.5 ml saline in test tube
  • Allow to stand for 10 minutes
  • Squeeze the swab dry against side of tube
  • Centrifuge saline at 2000 rpm for 10 minutes
  • Stain sediment with Diff Quik
  • > Detects 100% bitches mated within last 24 hours
  • > Detects 75% bitches mated within last 48 hours
  • Establish presence of sperm
  • Establish cytological state

If the risk of pregnancy was thought to be low apart from immediate administration of oestrogen or another product what else might be appropriate?

  • We think the risk is low but come and see us at day 21 or 28 and if she is pregnant we can terminate the pregnancy then
  • Prolactin inhibitor after day 20
  • Progesterone and prolactin together
  • Alizin any time during pregnancy- wait for diagnosis first?

Answers from wrap up:

  • If risk considered to be Low could scan at day 25

and treat if pregnant (NB > 50% will not be pregnant)

  • Spay
    • 3 months/ 65 days after oestrus so you know the bitch is in anoestrus at the time of spay
  • GnRH long acting implant
    • If he did not want her to come into oestrus again.
  • Doesn’t want her to be mated and doesn’t want the above= keep her away from dogs
