- MOA (RA) promote adenosine release (anti-inflammatory mediator)
- ADV GI intolerance, bone marrow suppression, liver cirrhosis

- MOA DEC lymphocyte proliferation by inhibiting dihydrooratoate dehydrogenase (DHODH)=suppress the de novo synthesis of pyrimidine nucleotides
- ADV long half life, so drug elimination is necessary for women who want baby
