Lithium Structure
Li+
Lithium MOA
- Inhibits phosphoinositide hydrolysis → (inositol depletion)
- Inhibits glycogen synthase kinase 3 (GSK-3) (↑BDNF, IGFs, WNTs expression)
- Uncouples G-proteins to adenylate cyclase (Vasopressin & TSH)
- Inhibits histone deacetylases - ↑ gene expression (substitutes for Na+)

- Thiazide diuretics & NSAIDs inhibit excretion and potentiate Li+ effects
