MOA PDE5I

A
  • Inhibits PDE5 which is in SM of corpus cavernosum and arteries of the lung.

in the penis; for erection, NO released, which causes CGMP to be produced and cause vasodilatation and engorgement.

PDE5 normally breaks down CGMP. so inhibit = increased conc

only with sexual stimulation

in lungs - pulm vasodilation similarly

  • hypotension, tachycardia and palpitations and small icnreased risk of vascular events
  • priapism - erection fails to subside
  • visual disorders if PDE6 is inhibited in retina
