DNA extraction - SB3 Flashcards
1
Q
what equipment is needed to extract DNA?
A
3g salt
100cm3 of water
beaker
10cm3 washing up liquid
50g of peas
water bath (60 degrees)
filter paper
funnel
flask
boiling tube
2 drops protease enzyme solution
ethanol (cold)
glass stirring rod
2
Q
what is the method for extracting DNA?
A
- dissolve salt in beaker filled with water - add washing up liquid and stir till salt is dissolved
- Mash peas - put peas into an empty beaker and add solution from 1 - stir slowly
- place beaker in water bath for 15 minutes
- Filter mixture and collect filtrate in beaker
- pour into boiling tube and add protease enzyme solution
- slowly pour in cold ethanol onto the side of the tube. equal amounts ethanol to filtrate
- Sit for few mins while precipitate forms between ethanol and filtrate
- DNA can be collected with stir rod
3
Q
why is DNA extraction used?
A
to identify origins of food and bases within
4
Q
why is washing up liquid important to the experiment?
A
Breaks down cell surface membranes and membranes around the nuclei
5
Q
why is ethanol used?
A
insoluble to DNA so guarantees precipitate is formed
