Indirect ELISA?
Antigen coated well, add specific antibody then add enzyme conjugated secondary antibody and add substrate and measure colour
Colour is proportional to the amount of a specific antibody
Sandwich Elisa?
Antibody coated well then add antigen to be measures, add enzyme conjugated secondary antibody then add substrate and measure colour
Colour proportional to the amount of antigen e.g cytokine, tumour marker
How does sandwich work?
AntiIgG then add plasma containing IgG then different anti igG binding on a different site

Antigen- human chorionic gonadotrophin
Why do you block with milk?
So there is specific binding of the antibody and the antibody doesn’t bind to the plastic
The typical shapes of a calibration curve is?
Sigmoid, because it might not be sensitive enough or there’s too much antibody
