Mechanism of action
A
Positive allosteric modulator for GABAa receptors->binds to benzo receptor site on chloride channels and potentiates binding and effects of GABA (increases Cl-concentration leading to hyperpolarization of the cell

Pharmacokinetics
A
- onset of action=3-5 minutes IV, 15 minutes IM
- Peak Effects= 20-60 minutes
- Duration of effects= <2 hours IV, 1-6 hours IM
- half life= 1-4 hours

Indications/field use
A
- Pre-medication sedation for cardioversion or TCP
- Post-Intubation sedation
- Seizure

Contraindications
A
- HYPOTENSION
- history of narrow angled glaucoma

Side effects/adverse reactions
A
Hypotension, drowsiness, amnesia, respiratory depression, AMS, blurred vision, N&V

Dose
A
Adult:
- Pre-Medication- 2-5mg, slow IV, IO, IM - Post Intubation- 2-5 mg q 5-10 minutes slow IV, IO - Seizure- 2-6mg IV, 10 mg IM, 10 IN
Pedi:
0.2mg/kg slow IV, max dose 10mg

Precautions
A
- May cause CNS and respiratory depression.
- Caution with acute alcohol intoxication
