Antiemetic

Mechanism of Action:
5-HT3 (serotonin) R antagonist ⇒ based on the theory that 5-HT3 receptors are present at the vagal nerve terminals and binding of serotonin initiates vomiting
reflex, Zofran would prevent this process.

- Onset of Action = Est. 10-30 minutes
- Peak Effects = Unknown (theory)

- Half-Life = 4 hours

Indications & Contraindications
Indications/Field Use:
• Nausea / Vomiting
Contraindications:
• Hypersensitivity

Side Effects/Adverse Reactions:
HA, dizziness, dry mouth
tachycardia, chest pain, prolonged QT interval
constipation, diarrhea

Adult & Pedi Dose:
(ADULT): 4mg slow IVP (IO) over 2-5 min
(PEDI): 0.1mg/kg slow IVP (IO) over 2-5 min

Precautions:
Lower risk of sedation and dystonia than other antiemetics

Drug-Drug Interactions:
May affect clearance and half-life with inducers and inhibitors of Cytochrome P-450 enzymes
(CYP3A4, CYP2D6, CYP1A2)
