cells are broken up and organelles are released and seperated out

cold to prevent denaturation of enzymes due to high temperature, buffered ph solution to ensure ph is constant as may effect enzyme activity and also sae water potential to prevent cell from bursting as a result of osmotic gain
what is a homogeniser
the cells are homeogenized which is like a blender it releases organneles from the cell the resultant fluid is called the homogenate as the whole cells are removed and large pieces of debris
the what is ultracentrifugation
the homogente is spun at high speed creating a centrifugal force which forces heaviest sediment at the bottom forming a pellet the remaining supernatant are removed and spun at an even faster speed
