- inhibits dihydrofolate reductase
- which is necessary for synthesis of purines and pyrimidines

- severe crohns disease
- moderate to severe active rheumatoid arthritis
- severe psoriasis unresponsive to conventional therapy

adverse effects

- anaemia
- decreased appetite
- increased risk of infection
