- Disrupt DNA synthesis
- Azathioprine: blocks purine synthesis mainly in lymphocytes
- Methotrexate: stops the action of the enzyme dihydrofolate needed for production of DNA

- Post transplantation immunosuppression
- Inflammatory bowel disease
- Renal vasculitis
- Paediatric leukaemia (methotrexate is used)
