Semen is diluted in ionic solution; Cryoprotectant added: glycerol most commonly used; Dilutes semen is cooled, sampled and frozen in liquid nitrogen at 196 degrees celcius

female sperm has 3-4% more DNA than male; relies on use of fluorescent dye that is taken up by DNA. Female determining DNA gives off more light; can be separated using cytometry
