what is DNA extraction
a process of purification of DNA from sample using a combination of physical and chemical methods
DNA first needs to be purified away from proteins and other cellular contaminants in order to be further used in testing
reason why people do this
genetic testing (if Newborn has a disease)
analyze forensic edvidence/ body identification
study a gene involved in cancer
4 main stages
- collect cheek dna
- burst cell pen to release dna
- separate dna from proteins and debris
- isolate concentrated dna
why is lysis solution added and what do its ingredients do?
separate and release DNA since it includes…
1. detergent: disrupts cell membrane and nucleur envelope to cause cell to burst open and release DNA.
- proteinase K: separates histones from the DNA
what does the concentrated salt do
will cause proteins and cellular debris to clump together
what does micro-centrifuge do? and where will dna be.
so protein and debris separate from liquid which contains the dna
- dna at top (liquid)
then after using isopropyl alcohol dna is not soluble to it so it comes out of the solution and 2nd time it would be at the bottom (pellets).
if doing this at home
can use fruits, chicken liver, vegetables, or anything living.
use blender to separate cells from one another
