What is the drug classification of acetaminophen?
Central analgesic; Non-narcotic analgesic, antipyretic
How is acetaminophen supplied?
120 to 325 mg tablets
Describe the MOA (pharmacodynamics) of acetaminophen.
- Inhibits prostaglandin synthesis through COX-2 inhibition in the CNS and to a lesser extent blocks pain impulses through peripheral action.
- Acts centrally on the hypothalamic heat regulating center, producing peripheral vasodilation (heat loss, skin erythema, diaphoresis)
What are the indications for acetaminophen use?
Relief of mild to moderate pain, fever
What is the ADULT dose for acetaminophen?
325 to 975 mg PO q4-6h ( daily max 4000mg)
1000mg IV q4-6h, max 4000mg daily
What is the pediatric dose for acetaminophen?
10 to 15mg/kg/ PO/PR 4-6 hours to max of 60mg/kg/day
List the contraindications to acetaminophen use.
- Hypersensitivity
- Severe hepatic impairment or severe active liver disease
What is a precaution for acetaminophen use?
Use with caution in hepatic impairment; renal impairment, alcohol dependency
Does acetaminophen cause histamine release?
Rarely
What happens if acetaminophen is given with benzodiazepines?
There is a synergistic effect. They can be used in combination to maintain “sedation” for advanced airways and advanced procedures.
Does acetaminophen have hypnotic activity?
Has little hypnotic activity
What is the toxic dose of acetaminophen?
150mg/kg
