Tests for Fungal Infections Flashcards
for certain bacteria & fungi that possess the enzyme Urease w/c hydrolases Urea releasing ammonia into the medium.
Urease test
This produces a change in the pH of the of the medium.
Urease test
Effect of urea to fungi
increases microbial antagonism w/c reduces its survival
Nitrate is transported from the medium into the cell cytosol where it is reduced to ammonium in 2 successive reactions catalyzed respectively by nitrate & nitrite reductase.
Nitrate assimilation in yeast
an agar that is made up & inoculated w/ a yeast & a carbohydrate. It is incubated from 1 - 3 days & even up to 24 days to allow the identification of yeast that are slow metabolizers.
Assimilation test for fungi:
is a polysaccharide predominant & specific constituent of fungal cell wall. It can be detected in serum during invasive fungal infections serving as a biomarker for Aspergillosis & invasive Candidiasis.
1 - 3 B - d - glucan
may be employed for the maintenance of stock culture of fungi, especially the black pigmented varieties. Addition of glucose will enhance the chromogenesis of Trichophyton rubrum.
Cornmeal Agar
is recommended for use in the cultivation of fungi & for the inducement of Chlamydospore formation by Candida albicans.
Tween 80 in Cornmeal agar
reduces surface tension & enhances the development of Chlamydospores.
Polysorbate 80
increases the level of a. acids, lipids, & organic acids in fungal cells. Helps to initiate alcoholic fermentation.
Thiamine
combines magnetic resonance w/ nanotechnology to identify whole Candida spp. cells.
T2 Magnetic Resonance assay
serves as enzymatic agent that breaks down debris in a specimen such as epithelial cells & WBC to view yeast or pseudohyphae. Freshly prepared is required because ___ can react w/ Carbon dioxide, giving wrong result.
KOH prep
can irritate the lungs when inhaled.
Hazards of KOH
with 2% glucose to support fungal growth & 0.5 ug/ml Methylene blue w/c enhances the zone edge definition.
Mueller - Hinton Agar for susceptibility testing
