What are antiulcer drugs used for?
Intended to counteract the processes that lead to GI ulcer formation
What can cause GI ulcer formation? (5)
- Hyperacidity
- Direct injury by drugs or chemicals
- Reflux of bile from duodenum into stomach
- Accumulation of metabolic toxins
- Decreased PGE production
What are most Antiulcer Drugs used as?
antacids in the stomach
How do antacids work?
decrease gastric irritation by decreasing acidity
What are some examples of systemic antacids? (3)
- cimetidine (Tagamet)
- ranitidine (Zantac)
- famotidine (Pepcid)
What are the general characteristics of systemic antacids? (2)
- decreases gastric acid secretion (HCl) in stomach by blocking histamine receptor cells
- Cimetidine inhibits metabolism of some other drugs by the liver
What are some examples of Non-systemic antacids? (3)
- calcium carbonate (Tums, Rolaids)
- magnesium hydroxide (Carmilax, Riopan)
- aluminum hydroxide (Amphojel)
What are the general characteristics of non-systemic antacids? (4)
- Minerals directly neutralize acid molecules in the stomach
- Ca and Al antacids can cause constipation
- Mg antacids can cause diarrhea
- May interfere with absorption of other drugs
What are the general characteristics of Omeprazole (Prilosec)? (2)
- Binds to an enzyme in the parietal cell to prevent secretion of H+ into the intestinal lumen
- HCl secretion
What are the general characteristics of Sucralfate (Carafate)? (3)
- Forms sticky paste that protects an active ulcer (helps heal ulcers)
- Promotes production of PGE
- Should not administer at the same time as antacid medications
What are the general characteristics of Misoprostol (Cytotec)? (2)
- synthetic PGE
