What are PCKS9 inhibtors? MOA?
A
- Monoclonal antibodies that reduce LDL receptor degradation in liver🡪increase LDL receptors🡪decrease circulating [LDL] (cholesterol)
- Ex. Evolocumab, Ex. Alirocumab—inactivate the enzyme=PCSK9 that causes LDL-receptor degration🡪increased LDL uptake from serum into cells
- Major effect: of all LDL-reducing drugs, PCKS9 inhibitors and STATINS have GREATEST ability to decrease LDL. Minor effect: increase HDL and decrease LDL
- Risk of myalgia, and CNS issues (delirium, dementia)
