CH 3 lecture Flashcards
a culture which contains a single
species of microorganism
PURE (axenic) CULTURE
increasing the population of
microorganisms by providing
their nutritional and physical
requirements
CULTIVATION
extracellular substances which provide the cell with materials for building protoplasm and for energy generation
NUTRIENTS
CULTURE MEDIUM uses (3)
- g___ and m___
- favor the p___ of particular compounds
- study m___ action
GROWTH AND MAINTENANCE
PRODUCTION

% of Solidifying Agent of:
1. Liquid
2. Semi-Solid
3. Solid
1.) 0
2.) 0.1 - 0.5
3.) 1.5 - 2.0
2 most used TYPES of SOLIDIFYING AGENT
Agar and Gelatin
TYPE of culture media acc to CHEMICAL COMPOSITION:
- all components are
chemically defined - NOT all components are
chemically defined
- Synthetic
- Complex
types of CULTURE MEDIA according to PURPOSE:
- distinguishes one type of bacteria from another
- used to determine qualitative/ quantitative production of such a
compound by an organism - used to increase the number of
microorganisms with unusual
physiological characteristics - can support most or almost all types of species
- with special reagents like pH indicators or dyes
- allows the growth of a specific
type of microorganism only
- Differential
- Assay
- Enrichment
- General Purpose
- Differential
- Selective
EXAMPLES of CULTURE MEDIA according to PURPOSE:
1. Blood Agar
2. Nutrient Agar
3. Eosin Methylene Blue Agar
4. Potato Dextrose Agar
5. Triple Sugar Iron Medium
6. Bacillus Cereus Agar
7. Fermentation Medium

TYPES of ISOLATION TECHNIQUES
- a macroscopically visible
growth or cluster of
microorganisms on a solid
medium - used if the desired microorganism is present at a higher level than
any other microorganism - isolation of specific types of
microorganisms by a
combination of nutrient and
physical conditions - for samples with low population: uses a sterile membrane filter having a pore size that retains microorganism
- uses a micropipette or a microprobe to physically pick a single cell and transfer it on an agar medium
- used for the isolation of unusual
physiological types of
microorganisms which are
present in small numbers and
which grow slowly
- Plating
- Serial Dilution
- Enrichment Culture
- Membrane Filter Technique
- Single-cell Isolation Technique
- Enrichment Culture
3 Types of PLATING
- Streak Plating
- Spread-Plate Method
- Pour-Plate Method
