cc lab finals Flashcards
containing compounds that are not proteins or polypeptides
nitrogen
Total NPN can be tested by making a
protein-free filtrate
Highest concentration of NPN in blood
Urea, 45%
Urea is synthesized in the
liver, from CO2 and ammonia, as the final product of amino acid catabolism.
is a nonspecific indicator of renal function
urea
States associated with elevated levels of urea in blood are referred to as
uremia & azotemia
Causes of urea plasma elevations
◼ Prerenal
◼ Renal
◼ and postrenal
is performed to differentiate between pre-renal and post-renal azotemia.
Parallel determination of urea and creatinine
Specimen for BUN
Serum and heparinized plasma
Enzymatic Method
◉ Nessler’s Method
◉ Berthelot Reaction
◉ Urease/GLDH Method
Urea is converted to ammonia by the enzyme urease.
NESSLER’S METHOD
Ammonia reacts with Nessler’s reagent giving rise to a
brown colored compound which is read at 450 nm.
Nessler’s reagent is composed of
potassium mercuric iodide.
Disadvantages of nessler method
Colour instability.
Turbidity.
Nonlinear calibration
