Objective lens

Chromatic aberration/colour fringing
Colour distortion creating an outline of unwanted colour along edges of an object
Dry mount
Solid specimen viewed whole or sectioned. Cover slip placed on top
Wet mount
Specimen suspended in liquid. Cover slip placed at angle.
Used for living organisms
Squash slides
Wet mount first prepared, lens tissue press down cover slip.
Smear slides
Edge of slide used to smear sample, creating a thin even coating of the sample. Cover slip then placed on top
Gram stain technique
2 types of bacteria, Gram-positive & Gram-negative. Crystal violet and iodine used to stain Gram-positive. Safranin dye used as counterstain.
Gram-positive is susceptible to penicillin.
Fixing
Method used to preserve a specimen in chemicals
Cell wall
Rigid structure, Provides support to the cell.
Plants- cellulose
Fungi- chitin
Prokaryotes- murein
Nucleus
Manages cell’s genetic material. Controlls growth and division
