QUININE: INDICATIONS
A
- Prevent night time cramps
- 1st line for Plasmodium Falciparum -MALARIA

QUININE: MOA
A
- Leg Cramps - reduce the excitability of the motor end plate in response to ACETYLCHOLINE STIMULATION (reduces frequency of muscle contractions)
- Malaria - Rapidly kills P.Falciparum parasites in schizont stage of the blood

QUININE: ADVERSE EFFECTS
A
- v toxic and can be fatal in OD
- Tinnitus, deafness + blindness (which may be permanent)
- GI upset, HS Rxns
- Prolongs the QT interval and causes a predisposition to Arrhythmias
- Hypoglycemia can also occur

QUININE: WARNINGS
A
- Existing hearing/visual loss
- Teratogenic -> should not be prescribed in the 1st trimester
- G6PD deficiency = Precipitate Haemolysis

QUININE:INTERACTIONS
A
- Drugs that prolong the QT interval -> can cause arrhythmias
- Amiodarone, antipsychotics, quinolones, macrolides, SSRIs
