HMG CoA Reductase Inhibitors (“statins”) Flashcards
1
Q
MOA
A
block HMG-CoA reductase = blocks cholesterol synthesis

Q
drug names to know
A
- Pravastatin
- Atorvastatin (Lipitor)
- Rosuvastatin (Crestor)
- Simvastatin
3
Q
common AE
A
- myalgia
- usually symmetrical, large proximal muscle groups
- less likely w lower doses
4
Q
why is myalgia less likely with pravastatin and rosuvastatin?
A
they are more hydrophilic
5
Q
what makes someone more likely to develop myalgia when taking statins?
A
- elderly
- female
- low BMI
- Asian descent
- excess alcohol
- high levels physical activity and trauma
- combo with interacting meds
6
Q
rare AE
A
- Myopathy
- Rhabdomyolysis
- ↑ risk with drug-drug interactions (especially fibrates)
7
Q
other AE
A
- dyspepsia (indigestion)
- headache
- ↑ liver function enzymes
- tendinopathy
- tendon rupture
8
Q
PT concerns
A
myositis, myalgia, persistent muscle effects, rhabdo, grapefruit juice inhibits CYPs, DDIs can cause increased statin conc.
