Controlled Ddel Overview W9 Flashcards
Why do we choose an oral route
Systemic delivery of a drug for many non-GI specific conditions
Also to treat GI conditions
This is the most acceptable form of medicine for adherence as wasted medicines cost the NHS millions every year
Challenges of oral route
Taste
Swallowing difficulties
Low bio availability
Dosing of oral drug delivery
The drug must be liberated at the correct rate and must escape degradation and metabolism as well as reaching sufficient concentrations in the target tissues q
When is the drug substance considered highly soluble?
When the highest dose strength is soluble in < 250 mL of water over a pH range of 1–7.5
When is a drug considered rapidly dissolving?
When > 85% of the amount dissolved within 30 minutes
The pH of the small intestine determines what?
The degree of ionisation and so controls the efficiency of absorption
Persoption
Special mode of permeation across the intestinal wall
What is uncontrolled release drug delivery?
No formulation constraint on the release of the drug from the delivery system
Conventional tablet or capsule
What is controlled release drug delivery?
There is a formulation constraint on the release of drug from delivery system
Constant/targeted release tablet or capsule (enteric coated)
What is a monolith device?Y
A block of material through which a drug is dispersed and released slowly
