Atherosclerosis 2 Flashcards
What do statins inhibit?
HMG-CoA Reductase
What do statins do?
Reduce circulating LDL chlesterol
What did Pravastatin do in the PRINCE trial?
Reduced CRP
What is protein prenylation involved in?
Leukocyte motility, activation proliferation and function
What do fibrates do?
Increae oxidation of free fatty acids and reduces hepatic synthesis triglycerides.
Increases expression of lipoprotein lipase.
Increases concentration of HDL cholesterol and expression of ApoA-1 and apo-AII
What are bile acid binding resins used for?
Reduction of LDL cholesterol
What do bile acids in the gut do?
Prevent enterohepatic circulation of bile acids
gain of function mutations found in PCSK9 leads to what?
Changes in LDL cholesterol levels
What is PCSK9?
A serine protease mainly expressed in the liver hepatocytes, intestine and kidneys
What does PCSK9 do?
Binds to LDL receptor and controls the level of protein
What do Evolocumab and aliorcumab do?
Target and inactivate PCSK9
What happens when PCSK9 is vound to LDL receptor?
Digested in lysosome
What is ezetimibe?
Cholesterol absorption inhibitor that targets uptake at the jejunal enterocyte brush border
What is the primary target of action for ezetimibe?
The cholesterol transport protein Nieman Pick C1 like 1 protein
