Why and where are are antibodies produced?
produced by B lymphocytes as part of the immune response in a vertebrate
What are antibodies?
Y-shaped globular proteins
What are specific antibodies specific to and why?
They are specific to antigens and bind with them to render them harmless
Where can antibodies be harvested from?
Serums in animal blood
Why do we harvest antibodies?
Allows us to detect and identify proteins from animals that have been exposed to certain antigens
What do we use to separate serum to identify its proteins?
Centrifugation
What is a monoclonal antibody?
An antibody with the same specificity as a stock of other antibodies and are used for immunoassay techniques
What is an immunoassay?
It’s a biochemical test that detects the presence of certain molecules via the use of an antibody or antigen
What two reporters help detect the presence of an antibody?
Chemiluminescence
Fluorescence
What is western blotting?
Technique used to identify proteins in a sample
What technique is used in western blotting?
SDS-page
Proteins blotted onto a membrane and then probed with fluorescent labelled monoclonal antibodies separating the protein
How do SDS-pages separate proteins?
By size alone as they are in a denatured state
Why do the molecules in SDS-pages go negative?
The gel with the SDS in them turns them negative
