- A tissue constituent is extracted from the body of X animal, and pureified.
- Injected into bloodstream of another animal. Where it works as an antigen and evokes an immune process, production og antibodies.
- The antibodies can be extracted from the blood, purified and characterized.

1.Collection of sample
Fixation:
Immersion
Perfusion through heart.

- Deep anaesthesia
- Cannule introduced through the left ventricle or into the aeorta
- wash out the blood with saline
- fix with paraformaldehyde og glutealdehyde
- remove wanted tissue or organ
- immersion fixed for some hours
- sectioning
