Topic 3 - Preparation of tissues for light microscopy Flashcards
1
Q
Process
A
- Excise and fix the tissue in fixative
- Remove water and replace with wa solvent (Ethanol)
- Embed the oriented specimen in molten wax (Parafin)
- Cut slices of the specimen after it is solid with a microtime and let them collect in a water bath
- Put slice onto slide.
- When its dry; remove the wax and rehydrate.
- When regydrated; stain(ex: hematoxylin eosin)
- Remove surplus stain and water, aply mounting medium and cover slip.
- Microscopic examination once mounting medium has set.
