NLS Flashcards
NLS
Nuclear localization sequence
NLS characteristics
not necessarily N-terminus; basic; not cleaved; importins and GTPase
a,b importins
responsible for recognizing NLS and translocating it to the nucleus
Ran GTP
responsible for removing a,b importins
CAS
GTPase activity; hydrolyses ran GTP and causes its translocation out of the nucleus
NTF2
responsible for bringing ran-gdp into the nucleus
ran-gef
phosphorylates Ran-GDP
NLS mode of action
1) a,b importins bind to NLS of protein and translocate it into the nucleus 2) Ran-GTP binds to a,b importins and causes their removal 3) CAS hydrolyzes ran GTP causing its translocation out of the nucleus 4) NFT2 binds ran-GDP bringing it back into the nucleus 5) Ran-GEF causes phosphorylation of Ran-GDP
