Core Practical- Enzymes On Initial Rate Of Reaction Flashcards
1
Q
Outline this experiment
A
Catalyse (in potatoes) to breakdown hydrogen peroxide into Oxygen and water. RoR can be identified by collecting volume of gas given off
2
Q
Why is only the initial rate of reaction used
A
Only time substrate conc is known, so constant across all enzyme concs
3
Q
Steps of experiment
A
1) cut 5mm thick potato discs
2) add different number of discs to boiling tubes
3) add 1ml of pH 7 buffer
4) place boiling tubes in 30 degree water bath
5) add 10ml of 1M hydrogen peroxide to tube
6) feed tube into upturned measuring cylinder and measure water displaced after 5 mins
