What are Light Microscopes?
light microscopes use light and lenses to form an image of a specimen and magnify it. Allow you to see individual and sub-cellular structures like the nuclei.
-Low magnification of up to 2000x
-Low resolving power (resolution) of below 0.3 micrometers
-lens made of glass
What are Electron Microscopes?
-uses beams of electrons to view specimens
-high magnification of up to 1,000,000x
-high resolving power of up to 0.0001 micrometers
-lens made of electromagnets
What is the equation for Magnification?
Magnification = Image Size/Actual Size
What is the method of preparing a cover slide?
1-Add a drop of water to a side
2-Place the specimen in the water
3-Place the edge of the coverslip on the slide so that it touches the edge of the water.
4-Slowly lower the coverslip to prevent forming and tapping air bubbles as they will obstruct the view.
How do you use a microscope (Light)?
1- clip the slide onto the stage
2-click lowest power objective lens (the one that produces the least magnification)
3-use coarse adjustment knob to just below the objective lens
4-look down at the eyepiece, use coarse knob to move the stage downward until image is roughly in focus
5-adjust focus with fine adjustment knob until image clear
6-if you need a greater magnification swap to a higher power objective lens + refocus
