What is the MoA of aspirin?
COX inhibitor
Aspirin irreversibly inhibits COX, reducing thromboxane A₂ production and platelet aggregation.
TXA2 usually gets released to go bind on another plt, cause change in conformation and plt degranulation
What is the indication for Aspirin?
Primary/secondary prevention of MI and stroke
Aspirin is used for both primary and secondary prevention of myocardial infarction (MI) and stroke.
What are the common side effects of Aspirin?
- GI bleeding
- Dyspepsia
- Increased bleeding risk
- Contraindicated in children (Reye syndrome)
What is the MoA of Clopidogrel (Plavix)?
ADP receptor antagonist
Clopidogrel irreversibly blocks the ADP receptor, reducing platelet activation and aggregation.
ADP receptor binding activates another GP2b/3a on membrane which is resp for crosslinking of plts w/fibrinogen
What is the primary use of Clopidogrel?
MI, stroke prevention, stent thrombosis
Clopidogrel is primarily used for prevention of myocardial infarction, stroke, and stent thrombosis.
What are the common side effects of Clopidogrel?
- Bleeding risk
- Neutropenia
- Possible drug interactions
What type of drug is Tirofiban (Aggrastat)?
GPIIb/IIIa antagonist.
Tirofiban inhibits the binding of fibrinogen to the GPIIb/IIIa receptor, preventing platelet aggregation.
What is the primary use of Tirofiban?
Acute coronary syndromes, percutaneous coronary interventions
