explain how a bight field microscope/ Kohler illumination manipulates light to be able to see it a specimen
light source is collected by a condenser which focus’ the light onto the specimen
the objective lens collects the light and directs it into the eyepiece able for us to see
what is phase contrast microscopy and why do we use it ?
uses light phase amplitude differences within microscopic objects
allows viewing of unstained specimens
what is Normarski/ differential interference contrast DIC
when is this used ?
uses polarised light with prisms to add a 3d shadow by adding additional contrast
used to investigate living cell non invasively and in real time
what is confocal microscopy ?
advantages/ disadvantages
uses a laser to excite florescent dye in a sample
then with this extremely precise image a computer creates a 3d reconstruction
advantages-
excellent resolutio for thick samples
collects light from a single focal point
disadvantages
photobleaching/ phototoxicity
increased sensitivity to noise
the method is labour intensive
