ability of drug to bind to receptor and produce drug receptor complex; stronger the affinity, more powerful the binding, and longer the duration (still relatively quickly unbind); agonist and antagonist possess affinity

once drug receptor complex forms, does it have ability to activate receptor and cause a response; only agonists possess affinity
