anti-fungals: differentiate between the drugs used to treat fungal infections
4 classifications of fungal infection based in terms of tissue/organ (become more dangerous)
superficial (outermost layer of skin), dermatophyte (skin, hair, nails), subcutaneous (innermost skin layers), systemic (primarily respiratory tract)
2 common categories of anti-fungals
azoles, polyenes
example of azole
fluconazole (treats candidasis)
example of polyene
amphotericin (treats fungal systemic infections)
mechanism of action of azoles
inhibit cytochrome P450-dependent enzymes involved in membrane sterol synthesis (e.g. conversion of lanosterol to ergosterol)
mechanism of action of polyenes
interact with cell membrane steroles (e.g. ergosterol) to produce membrane channels (pores)
