PCR made simple Flashcards
1
Q
WHat does pcr stand for
A
polymerase chain reaction
2
Q
polymerase chain reaction
A
- amplify a sample of DNA
3
Q
What is in the mixture of PCR
A
- reaction mesuture set up, contains:
- DNA
- free nucleotides
- primers
- DNA polyerase
4
Q
primer
A
- short pieces of DNA that are complementary to the bases at the start of the fragment you want
5
Q
DNA polymerase
A
enzyme that creates new DNA strands
6
Q
what are the strages of PCR
A
- heated to 95
- break H-bonds
- DNA polymerase doesnt denature
- mixture cooled to between 50 and 65
- so the primers can bind (anneal) to the strands
- heated to 72
- so DNA polymerase can work
- DNA polymerase lines up free DAN nucleotides alongisde each temp;late strand
- complementary base pairing means new complementary strands are formed
- wo new copies are formed and one cycle of PCR is complete
- repeat, all 4 strands used as templates
