MOUNTING Flashcards
refractive index of glass
1.518
mounting media which contains xylene
Resinous media
R.I. of DPX
1.532
R.I. of XAM

tree where Canada balsam is extracted
Abus balsamea
R.I. of Canada balsam
1.524
Resinous media for small tissue and dry rapidly
DPX
Recommended for whole mounts and is one of the best mounting media
Canada balsam
Mounting media for lipid which does not require xylene
Aqueous media
R.I. of water
1.33
Aqueous media for temporary mounting which has low refractive index
Water
Standard mounting medium for fat stains
Glycerin jelly
Glycerin jelly is also known as
Kaiser’s 1880
RI of glycerin jelly
1.47
Composition of glycerin jelly
- gelatin
- glycerol
- dH2O
- phenol crystals
Other name of Gum Arabic
Farrant’s
RI of Gum Arabic
1.43
Composition of Gum Arabic
- Gum Arabic
- dH2O
- glycerol
- sodium merthiolate
RI of apathy’s medium
1.52
Composition of apathy’s medium
- gum Arabic
- sucrose
- thymol
- dH2O
aqueous media for frozen sections
Brun’s fluid
other aqueous media
- permount
- HSR
- Clearmount
what is used to remove stain on skin
remove by using 0.5% acid alcohol –> tap water
how to restain old sections
shortcut:
“X - XhKhOhR”
- slide - xylene - remove coverslip - xylene - K2MnO4 - oxalic acid - restain
sealing the margins of the coverslip
ringing
purpose of ringing
- prevent escape/evaporation of fluid
- immobilize the coverslip
- prevent sticking of slides
what are the 2 adhesives used for ringing
a. Kronig cement
b. Durofix (cellulose adhesive)
what is the cellulose adhesive for ringing
Durofix
composition of Kronig cement
2 parts paraffin + 4-9 parts colophonium resin
purpose of mounting
- protect specimen from physical injury
- protect specimen from deterioration
- preserve for permanent keeping
- handle and store samples easier
Criteria for good mounting media
- RI must be as close as the RI of glass
- should not dry quickly
- should not dissolve or fade tissue
- should not cause shrinkage or distortion of tissue
- should be able to set hard for permanent mounting
