Methods - Lecture 9/9/21 Flashcards
Basophilic
Negatively charged tissue/structure (like nucleus) likes to bond with positive charged
Acidophilic
Positively charged tissue/structure has an affinity to acidic dyes
Hematoxylin
A basic dye, binds to neg structures
Eosin
An acidic dye, affinity to positively charged stuff
H and. E staining
Hematoxylin and eosin staining
Substances that stain poorly in H and E
Lipids and mucous
Lipid stains
Steatosis
Periodic-acid Schaffer (PAS)
Stains molecules rich in carbohydrates (GAGs, proteoglycans and mucus)
Silver stains
Used for reticular fibers and nerve cell processes (random nerve cells undergo rxn)
Masson’s trichrome
Three colored stain that distinguishes between cell and cytoplasm
Cytoplasm/keratin- red
Nuclei- dark/purple
Connective tissue- blue
Elastic fiber stain
Shows the elastin proteins in stretchy tissue
Immunohistochemistry
Uses antibody for specific structures
Examples of artifacts
- Cracks/tears
- Folds
- Sectioning defects
- Poor fixation
