Endocannabinoid System Flashcards
What is the endocannabinoid system responsible for?
Sleep and circadian rhythms
Analgesia
Positive mood
Energy balance
Stree coping
Are endocannabinoids synthesised on demand?
Yes
Levels of endocannabinoids are tightly regulated by which enzymes?
Synthetic and catabolic enzymes
Where are CB1 expressed?
Brain
Lungs
Muscles
GUT
Reproductive organs
Where are CB2 expressed?
Brain (microglia)
Tonsils
Skin
Bones
Spleen
Cannabinoid receptors-
What does delta9-THC mimic?
Mimics endocannabinoids and activates CB1 receptors
They are abundant in brain (cortex, hippocampus, stratum, mid brain) LOW IN BRAINSTEM
Where are CB1 receptors located?
Presynaptic
Are endocannabinoid synthesised on demand?
Yes on demand by LIPIDS
How does endocannabinoids reduce pain?
Endocannabinoida and CB receptors are found on pain pathways
Therefore CB1 and CB2 have anti-nociceptive effects
Paracetamol and NSAID effects on pain via CB receptors
The drugs are Inhibitors of FAAH which degrade anandamide
And
Increase endocannabinoids
Examples of endocannabinoid dysrefulstion in diseases
2
Reduced CB1 we expression in the brain
In
- Huntingtons disease
- Autism
