Describe the selectivity of muscarinic and nicotinic agonists (in general terms)
Muscarinic agonists are not ver selective between M1-5, some nicotinic agonists show selectivity between N1 and N2
What are the two major classes of cholinomimetic drugs?
Direct acting and indirect acting.
What are the two sub classes of direct acting cholinomimetic drugs
Choline esters and non-ester alkaloids
What effect does the addition of a beta methyl group have on acetylcholine?
reduces potency
What effect does the substitution of a carbamic acid group have on acetylcholine?
resistance to hydrolysis
What causes a reduction in peripheral resistance due to vasodilation when muscarinic agonists are given?
vasodilation is due to muscarinic receptor-induced NO (EDRF) production in endothelial cells causing relaxation of vascular smooth muscle.
How can muscarinic agonists reduce intraocular pressure?
By causing contraction of the ciliary body (M3)
How could you test to see if neostigmine dosage needed to be increased or decreased in myasthenia gravis?
Give a small dose of transiently acting edrophonium.
What does SLUDGE stand for?
Salivation, lacrimation, urination, defecation, GI distress, Emesis
