Tumour necrosis factor Flashcards
1
Q
What is TNF and what cell secretes it?
A
Pro-inflammatory cytokine secreted by macrophages
2
Q
Acts of TNF on the immune system
A
- activates macrophages and neutrophils
- costimulator for T cell activation
- key mediator of bodies response to Gram negative septicaemia
- anti-tumour effect (e.g. phospholipase activation)
3
Q
What receptors does TNF-alpha bind to?
A
p55 and p75 receptor (causes apoptosis)
4
Q
Systemic effects of TNF
A
pyrexia, increased acute phase proteins and disordered metabolism leading to cachexia
5
Q
give examples of TNF inhibitors
A
infliximab
etanercept
adalimumab
golimumab
6
Q
Adverse effects of TNF blockers
A
reactivation of latent TB
demyelination
