What is hematoxylin - pH, bind to what pH molecules, DNA/RNA color, why
basic - binds to acidic molecules - DNA/RNA appear blue/black = dark stain - acid molecules are basophilic
what is eosin - pH, color, binds to what molecules
acidic - red dye - basic molecules will bind to eosin - esocinophilic (acidophilic) - proteins stain with these - collagen is very eosinophilic
periodic acid Schiff (PAS) dyes
stain sacchrides
osmuim/sudan black
fixative = not stain! used to see lipids after H&E
can lipids be seen with stains?
No! they are washed out during dehydration with organic solvents
Masson’s trichrome
Most recipes produce red keratin and muscle fibers, blue or green collagen and bone, light red or pink cytoplasm, and dark brown to black cell nuclei.
Methylene blue
basic, blue good for staining specific granules in WBCs (azurophilic)
Romanovsky Stain
methylene blue + eosin for blood film prep!
