bisphosphonates (“-dronates”) - MOA

A

*pyrophosphate analogs
*bind hydroxyapatite in bone → inhibit osteoclast activity & PROMOTE OSTEOCLAST APOPTOSIS → lower net bone resorption / inhibition of bone breakdown
*used as first-line therapy to treat osteoporosis and other bone conditions

examples: alendronate, risedronate, zoledronate, ibandronate
