Lab 1 Flashcards
What is the function of the eyepiece?
connects lens to the body and allows proper alignment of light travel
What is the magnification of the ocular lens?
10x
What is the function of the ocular lens?
enhances details of the specimen and magnifies image
What is the 4x lens called?
Scanning lens
What is the 4x lens used for?
quickly surveying a sample
What is the 10x lens called?
Low power lens
What is the 10x lens used for?
Ideal for observing larger specimen details
What is the 40x lens called?
High power lens
What is the 40x lens used for?
examining fine details of a specimen
What is the 100x lens called?
Oil Immersion Lens
What is the 100x lens used for?
enhancing resolution with oil
What is the function of the objective lens?
gathers light from specimen and helps with magnifying image
How should order of objective lens be looked at?
4x to 10x to 40x to 100x
How do you change objective lens?
use the revolving nose piece
How do you clean oil immersion lens?
with lens paper and a little alcohol
How do you calculate total magnification?
objective lens x ocular lens
What is the function of the stage?
holds the slide
What is the function of the slide adjuster knob?
moves slides without touching it
What does the coarse focus knob do?
use for 4x and 10x
- brings specimen into general focus
What does fine focus knob do?
sharpens image and clarity
What is the relationship between magnification and visual field?
Inversely proportional
- decrease magnification = increase in visual field
- increase magnification = decrease in visual field
What is the relationship between magnification and depth of field?
Inversely proportional
- increase in magnification = decrease in depth
- decrease in magnification = increase in depth
What is the function of the condenser lens?
regulates brightness of light
What is the bright field setting good for?
dead bacteria
- light passes through
- colored specimens
What is the dark field setting good for?
live, unstained specimens
- light reflects off
What is the function of the iris diaphragm?
adjusts light intensity by changing size of aperture
- like the iris
How do you handle the microscope?
- 2 hands (one on arm, one on base)
- completely on table top
- take off cover and put it back in the cabinet
- plug in and turn on
How do you put away the microscope?
- turn off
- wrap cord nicely
- lower and center stage
- light to lowest setting
- 4x lens on
- put into cabinet with ocular lens facing inside
