- Nondisjunction (94%)
- Robertsonian translocation (onto chromosome 14) [5% or higher if parent carries translocation themselves]

Physical features of Down’s Syndrome
A
face:
- upslanting palpebral fissures
- epicanthic folds
- Brushfield spots in iris
- protruding tongue
- small low-set ears, round/flat face
- flat occiput
Other:
- single palmar crease
- pronounced gap between big and first toe
- hypotonia
- congenital heart defects
- duodenal atresia
- Hirschsprung’s disease

- endocardial cushion defect (AKA atrioventricular septal canal defects)

- Subfertility: males = always due to impaired spermatogenesis. Females = subfertile. + increased incidence of problems with pregnancy and labour
